miRNA 1st Strand cDNA Synthesis Kit (by tailing A)是适用于microRNA (miRNA)加尾法合成cDNA的专用试剂盒,只需一步操作,即可完成加尾反应和逆转录反应。本产品以miRNA或总RNA为模板,在Poly A polymerase (PAP)的作用下,在其3'末端加Poly (A),随后使用通用逆转录引物进行逆转录反应。本产品中,HiScript miRNA Enzyme Mix包含专为miRNA逆转录定向改造的逆转录酶和PAP,配以优化的缓冲体系,在具有高灵敏度的同时,较大限度降低miRNA前体的逆转录,保证后续结果稳定可靠。

2. 优异的逆转录性能

以HeLa miRNA为模板(100 ng/μl),使用 Vazyme #MR201 以及品牌相关产品,按照各自说明书推荐程序进行逆转录反应,将得到的cDNA分别在六种引物体系(T1/2/5/12/15/20)中进行qPCR定量分析。从图中可以看出,除两个体系中存在部分ΔCT<-0.5,其余均ΔCT>0.5或-0.5<ΔCT<0.5,说明Vazyme #MR201的逆转录效率优于同类产品。

3. 较高的逆转录特异性

以HeLa miRNA为模板(100 ng/μl),分别使用专为miRNA逆转录定向改造的逆转录酶(Vazyme #MR201)和未定向改造的逆转录酶(其余组分与Vazyme #MR201一致)进行逆转录反应,在相同条件下对逆转录反应得到的cDNA进行qPCR检测2个体系。可以看到,使用Vazyme #MR201逆转录miRNA后qPCR检测得到的溶解曲线单峰且峰形优异,特异性更高。
