使用KAPA2G快速热启动DNA聚合酶或者竞争者野生型Taq DNA聚合酶组成的热启动聚合酶通过多重PCR扩增人Duchenne肌肉失调基因的7个片段。反应物（25ul）包含100 ng（1，3和5道）或者10 ng（2，4和6道）女性人基因组DNA，0.25 uM各自的引物，3 mM Mg和一个单位的酶。KAPA2G快速热启动DNA聚合酶的反应在1.5倍浓度KAPA2G热启动缓冲液A中进行，竞争者的反应在1倍浓度反应缓冲液中进行。使用机器的型号是Eppendorf Mastercycler epgradient S，采用标准的3步法（30个循环），对于KAPA2G快速热启动DNA聚合酶和竞争者I，起始变性/酶恢复活性时间是2分钟，对于竞争者A是10分钟。对于KAPA2G快速热启动DNA聚合酶，每个循环由变性15秒，复性30秒和延伸10秒组成。对于竞争者，每个循环由变性30秒(95对)，复性30秒(55度)和延伸45秒(72度)组成.每种酶的总反应时间如图所示。

KAPA2G快速热启动DNA聚合酶基于新一代聚合酶，具有内在比野生型Taq DNA聚合酶和其它DNA聚合酶更快速合成DNA的能力。使用KAPA2G快速热启动DNA聚合酶的快速PCR程序基于延伸时间缩短，从而显著缩短了PCR循环的时间而不改变反应的表现。

使用KAPA2G快速热启动DNA聚合酶，对于小于1 kb的人基因组DNA片段，少到一个目标拷贝，总共1秒的延伸时间已经足够了。对于大于1 kb的扩增子，每个循环用15秒/kb的延伸时间。

KAPA2G快速热启动DNA聚合酶结合优化的缓冲液能够显著缩短延伸时间和带来优异的表现。野生型Taq DNA聚合酶组成的热启动聚合酶在延伸时间缩短的情况下表现不能令人满意，与之相反，KAPA2G快速热启动DNA聚合酶的独特性质使其在总反应时间缩短40 – 70%时依然能够有效扩增。
